Vertex Named Top Pick at BofA on Strong Kidney Drug Trial Data

Background

BofA Securities has reiterated its "Buy" rating and $667 price objective for Vertex Pharmaceuticals (VRTX), naming the drugmaker a "top pick" following the release of compelling new data for its experimental kidney disease treatment.

RAINIER Trial Shows Strong Efficacy

The endorsement follows the presentation of full results from the RAINIER trial for povetacicept in IgA nephropathy (IgAN), a chronic kidney disease. The data, presented at the American Society of Nephrology meeting and published in the New England Journal of Medicine, showed significant efficacy, according to the BofA note.

Key findings from the trial at week 36 include:

  • 42% of patients treated with povetacicept achieved the target for protein in urine (proteinuria) of below 0.5 g/g, a key marker of kidney health.
  • This compares favorably to just 6% of patients who received a placebo.
  • The drug also led to a 38% average reduction in the immune protein immunoglobulin G (IgG), which BofA noted compares well against reductions of 32% and 36% reported for rival therapies.

Importantly, BofA highlighted that no patients discontinued treatment due to their IgG levels falling below a critical threshold, suggesting a manageable safety profile.

Path to Market and Commercial Outlook

Investors are now focused on povetacicept's upcoming Prescription Drug User Fee Act (PDUFA) date of November 30, which is the deadline for the U.S. Food and Drug Administration to make an approval decision. BofA noted that Vertex management does not anticipate insurers will require patients to try other therapies first, a process known as step therapy.

Vertex management reportedly stated that approximately 70% of IgAN patients are commercially insured and that the company has been in discussions with major pharmacy benefit managers (PBMs) since July 2025. Coverage updates are anticipated in the first half of 2027. The company sees its kidney drug pipeline as a potential franchise worth approximately $10 billion.

Analyst View and Pipeline Catalysts

BofA's valuation model attributes $28 per share to povetacicept's potential in the IgAN indication alone. Looking ahead, the next major catalyst for Vertex's kidney franchise is an interim analysis of the AMPLITUDE trial for another drug candidate, inaxaplin, which is expected in early 2027.

While maintaining a positive outlook, the bank also cited potential downside risks for the company. These include potential pricing pushback from payers, challenges in securing reimbursement in the European Union, the inherent risk of clinical trial failures, and the emergence of new competitors in its core cystic fibrosis market.
