Background

Shares of Xenon Pharmaceuticals Inc. (NASDAQ:XENE) fell sharply in after-hours trading Thursday after the company announced it was voluntarily pausing new patient enrollment in clinical trials for its drug candidate azetukalner in psychiatric indications.

The stock plunged by as much as 16% following the news, which introduces uncertainty into a key area of the drug's development pipeline.

Details of the Trial Pause

Xenon stated the pause is a precautionary measure affecting its ongoing studies of azetukalner for major depressive disorder (MDD) and bipolar depression. The decision was made in consultation with the company's Data Safety Monitoring Board after an analysis of neuropsychiatric adverse events.

According to the company, the rate and severity of the observed events are consistent with the drug's known safety profile and mechanism. However, Xenon also noted that such adverse events had not been seen in its earlier Phase 2 X-NOVA study in MDD. Patients who are already enrolled in the psychiatry studies will continue treatment, and the company is evaluating potential changes to dosing to mitigate the side effects.

Epilepsy Program Unaffected

Crucially, the voluntary pause does not impact the company's development program for azetukalner in epilepsy. Xenon confirmed that its ongoing Phase 3 trials for focal seizures (X-TOLE3) and primary generalized tonic-clonic seizures (X-ACKT) will continue to enroll patients.

The company recently submitted a New Drug Application (NDA) to the U.S. Food and Drug Administration for azetukalner as a treatment for focal seizures, a significant milestone based on positive data from prior studies.

Market and Pipeline Impact

For investors, the halt clouds the timeline for azetukalner's potential expansion into the large psychiatric market. Despite the pause on new enrollment, Xenon said its X-NOVA2 study in MDD is now complete with approximately 360 patients, or about 80% of its initial target.

The company stated this patient count is sufficient to power the study to show a clinically meaningful change in its primary endpoint. Xenon still expects to release topline data from this MDD trial in the first quarter of 2027.
