Pyxis Oncology Shares Fall 7% Despite Positive Cancer Drug Trial Data

Shares of Pyxis Oncology (NASDAQ:PYXS) fell 7% in Tuesday's trading session despite the company reporting positive updated data from a Phase 1 study of its lead drug candidate. The market's reaction suggests investors may be weighing the promising efficacy results against the drug's safety profile and long path to potential approval.
Trial Shows Efficacy
Pyxis announced that its drug, Micvotabart Pelidotin (MICVO), demonstrated significant clinical activity in patients with recurrent or metastatic head and neck squamous cell carcinoma. The company reported the updated results from its monotherapy trial for patients who have already received other treatments.
Key efficacy findings from 33 evaluable patients include:
- A confirmed objective response rate (ORR) of 36%.
- A disease control rate (DCR) of 94%.
- A median progression-free survival of 6.2 months.
- A 12-month overall survival probability of 79%.
The company noted that the median overall survival has not yet been reached, which is often considered a positive signal in oncology trials. MICVO is an antibody-drug conjugate (ADC) that targets a structural component of the tumor's extracellular matrix.
Safety Profile and Market Reaction
AdDespite the positive efficacy data, the safety results may have tempered investor enthusiasm. According to the company, 54.3% of the 35 safety-evaluated patients experienced treatment-related adverse events of Grade 3 or higher. Furthermore, 14.3% of patients discontinued the treatment due to these adverse events.
While Pyxis stated there were no new safety signals and no treatment-related deaths, the rate of severe adverse events and discontinuations can be a key point of focus for investors evaluating a drug's overall risk-benefit profile. This type of "sell the news" reaction is not uncommon in the biotech sector, where trial results may not meet the market's highest expectations or when timelines for future development are lengthy.
Path to Pivotal Study
Pyxis Oncology outlined its next steps for the MICVO program, planning to meet with the U.S. Food and Drug Administration (FDA) in the first quarter of 2027. Following that meeting, the company intends to launch a pivotal Phase 3 study, named Headliner, in mid-2027.
The planned randomized study will enroll approximately 500 patients and compare MICVO against the investigator's choice of standard-of-care treatments, including cetuximab, docetaxel, or methotrexate.
