Background

Xenon Pharmaceuticals (NASDAQ: XENE) stock plunged 25.2% in after-hours trading following a dual announcement that paired a key regulatory achievement with an unexpected safety issue in its clinical pipeline.

Safety Concerns Halt Psychiatry Trials

The primary driver of the stock's decline was Xenon's decision to voluntarily pause new patient enrollment in its ongoing Phase 3 trials for azetukalner as a treatment for major depressive disorder (MDD) and bipolar depression. According to the company, the pause was a precautionary measure initiated after an analysis revealed neuropsychiatric adverse events in study participants.

While Xenon noted these events were consistent with the drug's mechanism of action, it also stated they had not been observed in the earlier Phase 2 X-NOVA trial for MDD. The company said the decision was made in consultation with its Data Safety Monitoring Board and that patients already enrolled in the trials will continue to receive treatment.

Epilepsy Filing Overshadowed

In the same announcement, Xenon confirmed it had submitted a New Drug Application (NDA) to the U.S. Food and Drug Administration (FDA) for azetukalner as a treatment for focal onset seizures in epilepsy. This filing was a long-anticipated milestone for the company, following positive results from its Phase 3 X-TOLE2 study.

However, the positive regulatory news was completely eclipsed by the negative safety signal from the psychiatry program. The market's severe reaction underscores the significant value investors had placed on azetukalner's potential in the larger psychiatric drug market.

Market Impact and Outlook

The sharp sell-off introduces material uncertainty into the timeline and ultimate viability of Xenon's psychiatry program. Topline data from the Phase 3 X-NOVA2 trial, previously anticipated in the first half of 2027, is now in question.

The stock's after-hours move to $42.90 erases months of gains and pushes it toward the lower end of its 52-week range of $35.97–$72.66. The move was specific to the company's news, as the broader S&P 500 and Nasdaq indices were both relatively flat during the session.
