Rocket Pharmaceuticals Shares Climb on FDA Agreement for Danon Disease Trial

Rocket Pharmaceuticals (NASDAQ: RCKT) shares rose by as much as 8% on Tuesday after the company announced a key alignment with the U.S. Food and Drug Administration (FDA) on its pivotal Phase 2 trial for RP-A501, a gene therapy candidate for Danon disease.
FDA Aligns on Trial Design
Rocket Pharmaceuticals reported that the FDA has agreed on the critical components of the trial intended to support a potential accelerated approval for RP-A501. This regulatory clarity is a significant milestone, as it provides a defined pathway for the drug's development program.
The agency confirmed the pivotal efficacy population will consist of 12 patients. It also endorsed the trial's co-primary endpoints, which will be assessed at 12 months post-treatment. The FDA's support includes continued enrollment and dosing at a recalibrated dose using commercial-grade product.
Trial Details and Safety Update
The pivotal study will enroll 12 male patients treated with RP-A501 at a dose of 3.8 × 10¹³ genome copies per kilogram. According to the company, the first three patients treated under a modified protocol will count toward this total, leaving nine additional patients to be enrolled.
AdRocket Pharmaceuticals expects to complete dosing for the remaining patients by mid-2027. The company also noted that the FDA reviewed initial safety data from the first three patients, who have completed at least four weeks of follow-up. No clinical evidence of thrombotic microangiopathy or capillary leak syndrome, which are potential side effects, was observed in these patients.
Context and Next Steps
Danon disease is a rare, inherited condition that weakens muscles, particularly the heart, and can lead to life-threatening heart failure. The positive regulatory feedback reduces uncertainty for the RP-A501 program, a key asset in Rocket's pipeline, which prompted the favorable investor reaction.
Rocket plans to provide a comprehensive update on its Danon disease program during a virtual investor webinar scheduled for October 6, 2026.
