Tyra Biosciences Shares Plunge After Bladder Cancer Drug Data Misses Analyst Expectations

Shares of Tyra Biosciences (NASDAQ: TYRA) plunged in pre-open trading after the company announced Phase 2 trial results for its oral cancer therapy, dabogratinib, that fell short of investor expectations. The stock was down 31.1% to $18.42 after the data readout raised concerns about the drug's competitive profile.
Trial Data Misses Key Benchmark
The sell-off was triggered by results from the SURF302 study, which evaluated dabogratinib in patients with a specific form of non-muscle invasive bladder cancer. While the drug achieved a 79% overall response rate, its 64% best overall complete response (CR) rate at the 60 mg dose was the focus for investors.
This figure was a clear disappointment, as analysts had established a higher bar for success. According to Investing.com, both Piper Sandler and H.C. Wainwright had publicly set a 70% three-month complete response rate as the minimum threshold, making the 64% result a significant miss against the consensus benchmark.
Despite the efficacy miss, the company reported a favorable safety profile for the drug. Most adverse events were Grade 1 or 2, with no severe Grade 4 or 5 events, and no patients at the 60 mg dose required dose reductions or discontinued treatment.
Market Reaction and Context
The sharp decline reflects several factors converging at once. The primary driver was the failure to clear a well-publicized efficacy hurdle for what Piper Sandler had previously estimated to be a potential greater-than-$1 billion market opportunity for dabogratinib.
AdFurthermore, Tyra Biosciences stock was richly valued heading into the data release, having surged approximately 13% in the prior week alone. This suggests the market had already priced in a more positive outcome, leaving the stock vulnerable to a significant correction on disappointing news.
The broader market environment provided little support, with major U.S. indices trading lower amid macroeconomic uncertainty, creating a risk-off sentiment for speculative biotechnology stocks.
Path Forward for Dabogratinib
Tyra Biosciences stated that it has identified the 60 mg once-daily dose to support its planned registrational development strategy. The company also indicated it would explore a 70 mg cohort to potentially improve efficacy.
However, for now, investors are aggressively repricing the stock to account for the heightened uncertainty around the drug's regulatory and commercial path following the below-benchmark trial results.
