Genmab Stock Surges After Head and Neck Cancer Drug Halves Death Risk in Phase 3 Trial

Shares of Genmab A/S (GMAB) surged more than 16% in morning trading after the Danish biotechnology company announced decisively positive results from a late-stage clinical trial for a new head and neck cancer treatment. The company reported its drug combination cut the risk of death by half, triggering a wave of investor optimism.
Pivotal Trial Results
The Phase 3 LiGeR-HN1 trial evaluated the combination of petosemtamab and pembrolizumab in patients with first-line PD-L1-positive recurrent or metastatic head and neck squamous cell carcinoma. According to the company's announcement, the study met its primary endpoint of overall survival with high statistical significance.
Key findings from the trial include:
- A 50% reduction in the risk of death compared to treatment with pembrolizumab alone, demonstrated by a hazard ratio of 0.50 (p-value < 0.0001).
- A statistically significant improvement in objective response rate, with an odds ratio of 3.17.
- A safety profile that was reported as generally well-tolerated and consistent with the known profiles of the individual drugs, with no new safety signals identified.
AdEverett E. Vokes, Chair of the Department of Medicine at University of Chicago Medicine, commented on the data, calling it "the first positive Phase 3 study in this patient population in several years" and a "major milestone with the potential to redefine care." Genmab stated it looks forward to engaging with regulatory authorities globally.
Market Reaction and Analyst Context
In response to the news, Genmab's American Depositary Receipts (ADRs) rallied 16.8%, reaching a new 52-week high of $52.85 during intraday trading. The stock's performance was a clear outlier, as the broader S&P 500 was up just 0.1% and the Nasdaq was nearly flat, underscoring that the rally was driven by company-specific catalysts.
The positive clinical data followed recent bullish calls from Wall Street analysts. On October 27, JPMorgan upgraded Genmab to Overweight from Neutral and raised its price target to DKK 3,500. On the same day, TD Cowen maintained its Buy rating and increased its price objective on the stock from $43.00 to $52.00, signaling growing institutional conviction ahead of the announcement.
