Foghorn Therapeutics Stock Plummets After Discontinuing Lead Cancer Drug, Ending Eli Lilly Partnership

Summary
Foghorn Therapeutics (FHTX) shares plunged after the company and partner Eli Lilly discontinued their lead oncology asset following a failed clinical trial. The company also announced a major restructuring, including a 40% workforce reduction, to extend its cash runway.
Shares of Foghorn Therapeutics (NASDAQ: FHTX) plummeted 43.7% in pre-open trading after the company and its partner Eli Lilly announced they were discontinuing their lead oncology drug candidate, FHD-909. The decision follows a Phase 1 trial that failed to show sufficient efficacy, leading to the termination of all collaboration activities between the two companies.
Clinical Setback and Partnership Termination
The joint decision to halt development of FHD-909 was made after a Phase 1 dose escalation trial did not meet the efficacy threshold required to advance the program. In a statement, Foghorn and Lilly also confirmed the termination of a separate Selective SMARCA2 degrader program, effectively ending their partnership.
CEO Adrian Gottschalk acknowledged the disappointing outcome, stating that "the biology of the SMARCA2/4 synthetic lethality relationship has not translated into the level of efficacy required to further advance the program." He noted, however, that the drug candidate did demonstrate selective target engagement and had a favorable safety profile.
Strategic Restructuring and Pipeline Shift
In response to the clinical failure, Foghorn's Board of Directors approved a strategic reprioritization on September 30, 2026, which includes a significant organizational restructuring. The company will reduce its workforce by approximately 40%, leaving it with around 65 full-time employees. Foghorn expects to incur about $2.3 million in one-time charges in the fourth quarter of 2026 related to the layoffs.
AdThe company will now redirect resources to advance its wholly owned pipeline, which includes:
- A selective EP300 degrader
- A selective CBP degrader
- A novel oral immunology and inflammation program
- Its induced proximity platform
This strategic shift is expected to extend Foghorn's cash runway into the second half of 2029.
Market Impact
The sharp decline in Foghorn's stock was a company-specific event, occurring while broader U.S. equity indices were modestly positive. The news represents a major blow to investors, as FHD-909 was the company's most advanced clinical program and the foundation of its high-profile collaboration with a major pharmaceutical firm. The combination of the clinical failure, the complete dissolution of the Lilly partnership, and the subsequent workforce reduction prompted a significant reassessment of the company's near-term valuation.
